MCW-led Trial Evaluates Antibody Drug Conjugate for Endometrial Cancer
The MCW Cancer Center activated a phase 2 investigator-initiated trial led by William Bradley, MD, Professor of Gynecology and Oncology, that evaluates the safety and efficacy of an antibody drug conjugate in patients with advanced endometrial (uterine) cancer that has worsened after previous treatment.
The drug, enfortumab vedotin (EV), combines an antibody that binds to the protein Nectin 4 with a payload that disrupts cell division. EV is already used to treat urothelial cancer and may provide a durable, lower toxicity option for patients with advanced endometrial cancer.
“Recurrent endometrial cancer has no curative options, so improvements in disease control are much needed,” Dr. Bradley said.
The team plans to enroll 12 adult female patients at Froedtert Hospital with advanced or metastatic endometrial carcinoma who have progressed after prior chemotherapy and anti-PD-1 or another immunotherapy. Treatment will continue for up to one year and patients will be followed over time to monitor their health and response. If the EV is successful, the team will look at other treatment combinations.
The researchers will also work with partners in basic science to compare blood samples of patients who respond well to EV with those whose responses are not as robust, Dr. Bradley said.
